New reactive group for bioconjugation of DNA

A new reactive group for bioconjugation of DNA has been developed by the Hocek Lab and reported recently in Angewandte Chemie. 

Novel squaramate-linked nucleotide was designed, synthesized, and used as a building block for enzymatic synthesis of modified DNA which then undergoes selective cross-linking with Lys-containing peptides and proteins under physiological conditions. 

The new reactive group and cross-linking reaction extends the portfolio of methods for synthesis of DNA-protein conjugates and has the potential for applications in identification and study of DNA-binding proteins.
