Abstract (Preferential Targeting of Tenofovir to the Lymphatic System)
A novel lymphatic targeting isopropylalaninyl monoamidate prodrug of tenofovir (GS 7340) was prepared, its in vitro activity against HIV measured, and its pharmacokinetics in dogs characterized. The oral administration of GS 7340 to dogs results in enhanced levels of tenofovir diphosphate in PBMCs and other lymphatic tissues. Based on these preclinical data, GS 7340 was taken into a 14-day viral dynamic study in drug naïve HIV patients. Results will be discussed.
